General description

Exosome complex exonuclease RRP44 or DIS3 mitotic control homolog gene is mapped to human chromosome 13q21.33 and is expressed in two isoforms. DIS3 is present mainly in nuclear fraction. It comprises endoribonucleolytic (PIN) and the 3′-5′ exoribonucleolyitic (RNB) domains and belongs to the RNase D family.

Application

Anti-DIS3 antibody produced in rabbit may be used in immunoblotting and immunofluorescence.

Biochem/physiol Actions

The exosome is a multi-subunit complex important for the 3′-5′ processing and degradation of many types of RNAs, including mRNA, rRNA, snRNA, snoRNA, and tRNA. DIS3 and DIS3L are active exonucleases. DIS3 mitotic control homolog and EXOSC11, is a catalytic component of the eukaryotic RNA exosome. It displays exoribonuclease and endonuclease activity.. DIS3 is a tumor suppressor and mutations in the gene is implicated in plasma cell dyscrasias. It is also involved in the mRNA quality control, RNA processing and metabolic pathways.RNA metabolism including mRNA quality control regulation of gene expressionand small RNA processing Mutation in the domain region of DIS3 is implicated in multiple myeloma (MM).

Physical form

Solution in 0.01 M phosphate buffered saline pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8°C for up to one month. For extended storage freeze in working aliquots. Repeated freezing and thawing is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
