Description

General description

Huntingtin-interacting protein-1 (HIP1) is a cytosolic protein, which belongs to the huntingtin-interacting protein family. The HIP1 gene is expressed in the endothelium of small blood vessels, gastric epithelium, cardiac muscle, brain tissues and liver. HIP1 and HIP1-related (HIP1R) proteins share the same domain structure, comprising a phospholipid-binding epsin N-terminal homology (ENTH), a leucine zipper motif, a central coiled-coiled and a C-terminal actin-binding domain (THATCH). The gene HIP1 is mapped to human chromosome location 7q11.23.

Immunogen

synthetic peptide corresponding to a sequence at the C-terminus of human HIP1, conjugated to KLH. The corresponding sequence is identical in rat and mouse HIP1.

Application

Anti-HIP1 antibody produced in rabbit may be used in immunoblotting and immunoprecipitation.

Biochem/physiol Actions

Huntingtin-interacting protein-1 (HIP1) and its homolog HIP1-related (HIP1R) acts as a cofactor in clathrin-mediated membrane traffic and endocytosis. It is implicated in the early recruitment and late-stage maturation of coated pits vesicles. The protein binds to clathrin and the endocytic adaptor complex (AP2) and regulates the binding of HIP1 to actin, leading to alterations in the actin cytoskeleton structure. In addition to its role in endocytosis, HIP1 gene is highly expressed in a variety of human cancers, including colon and prostate cancer. It is also found in the brain, melanoma, breast and ovarian tumors. Overexpression of HIP1 transforms fibroblasts and is associated with increased levels of multiple growth factor receptors. The protein HIP1 is necessary for the survival of germline progenitors and several cell lines. The protein interacts with huntingtin (Htt) and is implicated in the pathogenesis of Huntington′s disease (HD). The fusion between HIP1 and platelet-derived growth factor β receptor is associated with chronic myelomonocytic leukemia.

Physical form

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers,is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
