General description

Monoclonal Anti-Rab25 (mouse IgG2a isotype) is derived from the hybridoma RAB25-20 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with a synthetic peptide. Ras-related protein (Rab25) is a member of the rat sarcoma (RAS) family of small guanosine triphosphatases (GTPases) restricted to an epithelial distribution. It is also expressed in gastrointestinal mucosa, lung and kidney. The Rab25 gene is located at the human chromosome location 1q22. It consists of a guanidine triphosphatase (GTPase) tertiary structure, a central barrel composed of a six-stranded β-sheet surrounded by α-helices.

Specificity

Monoclonal Anti-Rab25 recognizes human Rab25.

Immunogen

synthetic peptide corresponding to an internal region of human Rab25, conjugated to KLH. The corresponding sequence is identical in dog and monkey, and differs by a single amino acid in mouse, rat, pig and bovine Rab25.

Monoclonal Anti-Rab25 antibody produced in mouse may be used in immunoblotting and immunoprecipitation.

Biochem/physiol Actions

Ras-related protein (Rab25) is linked with the apical recycling system of epithelial cells. It is involved in the development and aggressiveness of ovarian and breast cancers. Rab25 plays a vital role in endosomal transport, recycling of cell-surface receptors and signaling proteins. It enhances oncogenic functions by regulating integrin recycling and intracellular signaling pathways. It also functions as a tumor suppressor gene in colorectal cancer, head and neck cancer.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8°C for up to one month. For extended storage, freeze at -20oC in working aliquots. Repeated freezing and thawing,or storage in “frost-free” freezers,is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
