Osteoclast-stimulating factor (OSF or OSTF1) is a small intracellular protein with a N-terminal proline-rich region, SRC Homology 3 (SH3) domain, three ankyrin repeats and a C-terminal aspartate-rich region. The OSF gene is mapped to human chromosome 9q21.13. OSF is expressed in osteoclast (OCL) as well as motor neurons. Structurally, OCF protein takes up a β-barrel fold.
Specificity
Anti-OSTF1 (N-terminal) recognizes human OSTF1.
Immunogen
synthetic peptide corresponding to the N-terminal region of human OSTF1, conjugated to KLH. The corresponding sequence is identical in rat and mouse.
Application
Anti-OSTF1 (N-terminal) antibody produced in rabbit may be used in immunoblotting and immunoprecipitation.
Biochem/physiol Actions
Osteoclast-stimulating factor (OSF or OSTF1) SRC Homology 3 (SH3) domain undergoes conformational changes to bind to proline-rich ligand. The C-terminal ankyrin repeats preferably binds to negative-charged binding partners. OSF plays an indirect role in osteoclast (OCL) formation as well as bone resorption. It favors OCL formation by interacting with sarcoma proto oncogene (c-Src) and its associated protein-based signaling pathway. OSF is implicated in the pathophysiology of spinal muscular atrophy by its possible interaction with survival motor neuron (SMN) proteins through the SH3 domain. OSF-SH3 domain interaction stimulates OCL formation and its inhibition may reduce bone resorption by OCL. A compromised bone resorption functionality in OCL further triggers bone loss and may lead to various bone disorders, including hyperparathyroidism, Paget′s disease, osteoporosis, estrogen deficiency, periodontal disease as well as inflammatory bone loss and bone malignancy.
Physical form
Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
Storage and Stability
For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers,is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
