General description

α-1,2-Mannosidase IB (MAN1B), also known as mannosidase α, class 1A, member 2 (MAN1A2), is a Golgi resident transmembrane protein that belongs to the glycosyl hydrolase family 47. MAN1B is a type II membrane protein containing 35 amino acids in the cytoplasmic region, a Ca2+ binding consensus sequence, and a single N-glycosylation site. MAN1B is highly conserved among the eukaryotes and the murine MAN1B protein is localized to the Golgi apparatus. The MAN1B gene is mapped to human chromosome 1p12.

Immunogen

synthetic peptide corresponding to an internal sequence within human MAN1A2, conjugated to KLH. The corresponding sequence is identical in mouse, rat, monkey, bovine and canine.

Anti-MAN1B /MAN1A2 antibody produced in rabbit is suitable for immunoblotting.

Biochem/physiol Actions

α-Mannosidases function at different stages of N-glycan maturation in mammalian cells. α-1,2-Mannosidase IB (MAN1B) progressively trims the α-1,2-linked mannose residues from Man9GlcNAc2, to form the processing intermediate Man5GlcNAc2. MAN1B is responsible for protein transport to the Golgi apparatus for processing. This gene might serve as a prognostic indicator in B cell lymphoma.

Physical form

Solution in 0.01 M phosphate buffered saline pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For extended storage freeze in working aliquots. Repeated freezing and thawing is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
