General description

Monoclonal Anti-DFF45 (mouse IgG1 isotype) is derived from the hybridoma 3F12 produced by the fusion of mouse myeloma cells and splenocytes from immunized BALB/c mice.
The gene DNA fragmentation factor subunit α (DFFA) is mapped to human chromosome 1p36.2. The encoded protein localizes to the chromatin fraction. It exists in association with DFF40, thereby forming a heterodimer. DFFA is popularly called as DFF45 (DNA fragmentation factor 45).

Specificity

Monoclonal Anti-DFF45 recognizes human DFF45 isoform 1 and isoform 2.

Biochem/physiol Actions

DNA fragmentation factor subunit alpha (DFFA) is a component of DNA fragmentation factor (DFF). It is mainly associated with apoptosis and genomic stability. Caspase-3-mediated cleavage of DFFA releases DFF40 (DNA fragmentation factor 40) which degrades chromosomal DNA. During menstruation, the apoptosis of endometrium cells is associated with DFFA. Its expression decreases after menopause. Silencing of DFFA increases doxorubicin effects in breast cancer cells.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

Store at -20 oC. For continuous use,the product may be stored at 2-8oCfor up to one month. For extended storage, freeze at -20oC in working aliquots. Repeated freezing and thawing,or storage in “frost-free” freezers,is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discardedif not used within 12 hours.
