General description

Monoclonal Anti-Importin 4 (rat IgG2a isotype) is derived from the hybridoma 3C2 produced by the fusion of mouse myeloma cells and splenocytes from immunized rat.

Specificity

Monoclonal Anti-importin 4 recognizes human, mouse, rat, monkey and hamster importin 4.

Immunogen

mouse importin 4 fusion protein.

Application

Monoclonal Anti-Importin 4 antibody produced in rat is suitable for immunoblotting and immunocytochemistry.

Biochem/physiol Actions

Importin-4 is involved in the nuclear import of ribosomal protein S3a (rpS3a), as well as vitamin D receptor (VDR). Furthermore, importin-4 has been found to mediate the nuclear import of human cytomegalovirus UL84 by recognizing a non-classical nuclear localization signal (NLS). Importin-4 is also involved in transporting transition protein 2 (TP2) and interacts with the NLS sequence of TP2 in the spermatid. A three-dimensional (3D) structure of importin-4 indicates that the three helices (helices 21 to 23) of the importin-4 protein contribute to the NLS of TP2 binding pocket.

Physical form

Solution in 0.01M phosphate buffered saline pH 7.4, containing 15 mM sodium azide.

Storage and Stability

Store at -20 °C. For continuous use, the product may be stored at 2-8°C for up to one month. For extended storage freeze at -20 °Cinworking aliquots. Repeated freezing and thawing,or storage in “frost-free” freezers,is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.

Mammalian spermiogenesis is characterized by a unique chromatin-remodeling process in which histones are replaced by transition protein 1 (TP1), TP2, and TP4, which are further replaced by protamines. We showed previously that the import of TP2 into the haploid spermatid
