General description

Protein phosphatase, Mg2+/Mn2+ dependent 1M (PPM1M), also known as protein phosphatase 2C isoform η (PP2C-η or PP2CE) is a novel member of the protein serine/threonine phosphatase 2C family. At least 14 distinct PP2C family members have been identified in mammalian cells, all sharing six conserved structural motifs. PPM1M is ubiquitously expressed and localizes in the nucleus of cells. The PPM1M gene is mapped to human chromosome 3p21.2.

Specificity

Anti-PPM1M (C-terminal) recognizes human PPM1M.

Application

Anti-PPM1M (C-terminal) antibody is suitable for use in immunoprecipitation (2.5-5.0 μg) using lysates of HEK-293T cells over-expressing human PPM1M. The product may also be used for western blot (approx. 33 kDa).
Anti-PPM1M (C-terminal) antibody produced in rabbit is suitable for immunoblotting and immunoprecipitation.

Biochem/physiol Actions

Decreased PPM1M gene expression is associated with the inhibition of Herpes simplex virus 1 (HSV-1) infection.
Dephosphorylation of inhibitor of nuclear factor κ B kinase subunit β (IKKβ) by protein phosphatase, Mg2+/Mn2+ dependent 1M (PPM1M) inhibits interleukin-1 (IL-1)-nuclear factor κB (NF-κB) mediated signal transduction. Thus, PPM1M may have implications in regulating pro-inflammatory responses.

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide

Storage and Stability

Store at –20 °C. For continuous use, store at 2–8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
