General description

PU.1 is a transcription factor that belongs to the erythroblast transformation specific (ETS) family. This protein is highly expressed in hematopoietic cells like mast cells, B lymphocytes, neutrophils, early erythroblasts, and macrophages. PU.1 protein consists of a highly conserved DNA binding domain, a glutamine-rich domain, acidic residues towards the N-terminal, and a central proline-glutamic acid-serine-threonine (PEST) domain.

Immunogen

Recombinant protein encompassing a sequence within the center region of zebrafish PU.1/Spi1.

Application

Suggested starting dilutions are as follows: IHC-Wm: 1:100-1:500. Not yet tested in other applications. Optimal working dilutions should be determined experimentally by the end user.

Biochem/physiol Actions

PU.1 transcription factor facilitates the regulation of myeloid differentiation. This transcription factor is also involved in regulating several other myeloid genes such as macrophage colony-stimulating factor (M-CSF) receptor, CD11 antigen-like family member B (CD11B), granulocyte colony-stimulating factor (G-CSF) receptor, and granulocyte-macrophage colony-stimulating factor (GM-CSF) receptor. Dysregulation of the PU.1 gene results in an increase in erythroblast proliferation, inhibits erythroid differentiation and leads to erythroleukemia.
