Gene Information

human ... THPO(7066)

General description

Megakaryocytopoiesis is the cellular development process that leads to platelet production. THPO is a humoral growth factor that is necessary for megakaryocyte proliferation and maturation, as well as for thrombopoiesis. It is the ligand for MLP/C_MPL, the product of myeloproliferative leukemia virus oncogene. It is a lineage specific growth factor, produced in the liver, kidney and skeletal muscle. It is an acidic glycoprotein. The gene encoding it is localized on human chromosome 3q27.1.

Immunogen

THPO (NP_000451, 305-339)
This antibody is generated from rabbits immunized with a KLH conjugated synthetic peptide at the C-term of human THPO.

Biochem/physiol Actions

Thrombopoietin (TPO) stimulates the proliferation and maturation of megakaryocytes, and promotes increased circulating levels of platelets in vivo. TPO signals through the cellular proto-oncogene (c-mpl) receptor and acts as an important regulator of circulating platelets. High levels of TPO have been observed in patients with aplastic anemia. Human and murine TPO exhibits cross-species reactivity.

Physical form

Purified polyclonal antibody supplied in PBS with 0.09% (W/V) sodium azide.

We recently identified 2 siblings afflicted with idiopathic, autosomal recessive aplastic anemia. Whole-exome sequencing identified a novel homozygous missense mutation in thrombopoietin (THPO, c.112C>T) in both affected siblings. This mutation encodes an arginine to cysteine substitution at residue 38 or
