General description

Monoclonal Anti-Syntaxin 6 (mouse IgG1 isotype) is derived from the 3D10 hybridoma produced by the fusion of NS-1 mouse myeloma cells and splenocytes from BALB/c mice. The syntaxin 6 (STX6) gene codes for a 255 amino acid protein. The protein is expressed at high level in brain, lung and kidney. STX6 has two regions, such as H1 and H2 domains. The H2 domain confers trans-Golgi localization. It belongs to the syntaxin family. STX6 gene is mainly expressed in endosomes and trans-Golgi network and is located on human chromosome 1q25.3.

Monoclonal Anti-Syntaxin 6 recognizes rat, mouse, and human Syntaxin 6 (approx. 29 kDa).

Biochem/physiol Actions

Syntaxin 6 (STX6) protein participates in vesicular trafficking between the trans-Golgi network and endosomes or lysosomes. It interacts with the mammalian homolog of vacuolar protein sorting-associated protein 45 (VPS45), a sec1-like protein implicated in Golgi for prevacuolar compartment trafficking. STX6 plays a key role in controlling the caveolae-dependent endocytosis. It is actively involved in the maturation of endosome and GM1 (a ganglioside with one sialic acid) ganglioside membrane delivery. STX6 also plays a key role in reinternalization of glucose transporter-4 (Glu4) and sub endosomal Glu4 sorting. Overexpression of STX6 gene is observed in breast, tongue, colon, pancreatic, liver, prostate, skin, lung, cervical and gastric cancers. Antibodies, which react specifically with the syntaxin 6 protein are considered as an essential tool for studying vesicle trafficking in the cell.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

Store at 2-8 °C for up to one month. For prolonged storage, freeze in working aliquots at −20 °C. Repeated freezing and thawing is not recommended. Storage in frost-free freezers is also not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
