Human Sin3A is a 1273 amino acids protein containing paired amphipathic helix (PAH) domains, important for protein-protein interactions.
SIN3A (switch-insensitive 3 family member A) is a transcriptional repressor and MeCP2 (methyl-CpG binding protein 2) interactor. It is located on human chromosome 15q24. The product encoded by SIN3A gene is a constituent of the histone deacetylase (HDAC) complex.
Immunogen
synthetic peptide corresponding to amino acids 1267-1282 of human Sin3A, conjugated to KLH via an N-terminal added cysteine residue. The immunizing peptide is conserved in human, mouse, and rat and is not present in Sin3B.
Application
Anti-Sin3A, C-Terminal antibody produced in rabbit has been used in immunoblotting and immunoprecipitation.
Biochem/physiol Actions
SIN3A (switch-insensitive 3 family member A) along Sin3B is essential for transcription and growth suppressor functions of the mitotic spindle assembly checkpoint protein (Mad1) and interferon-induced GTP-binding protein (Mx1). It also controls cellular functions like differentiation, oncogenesis, apoptosis and cell fate determination. The Sin3A/HDAC corepressor comprises Sin3A along with histone deacetylases (HDAC1 and 2), pRB-associated proteins (RbAp46, RbAp48) and Sin3A Associated Protein 30 h(SAP30). Repression of transcription by the Sin3A/HDAC complex interaction with O-GlcNAc transferase (OGT) leads to the inactivation of transcription factors and RNA Polymerase II.
SIN3A (switch-insensitive 3 family member A) plays an important role in the growth of early embryos. It helps in the multiplication and survival of various cells by suppressing diverse signaling pathways. It acts as a transcriptional regulator for the formation of cortical brain. Absence of Sin3A can suppress the development of breast cancer cell by elevating apoptosis.
