General description

Rho GTPases belong to the Ras superfamily of small GTPases, which consists of the Rho, small GTP binding protein (Rac) and cell division cycle 42 (Cdc42) subgroups. Ras homolog family member B (RhoB) is mapped to human chromosome 2p24.1. It is present in the endosomal compartment. The farnesylated form present in multivesicular late endosomes and geranylgeranylated form localizes in plasma membrane. The C-terminal hypervariable domain contains the CAAX box prenylation motif, a polybasic Lys-rich domain, and may contain additional Cys residues required for palmitoylation.

Immunogen

synthetic peptide corresponding to amino acids 177-188 located near the C-terminus of human RhoB.

Application

Anti-RhoB antibody produced in rabbit has been used in immunoblotting.

Biochem/physiol Actions

Ras Homolog Family Member B (RhoB) modulates Rac1 trafficking and may play a key role in maintaining the integrity of endothelial cells during inflammation. It functions as tumor suppressor and its absence is implicated in tumors like cervical, breast cancer and clear cell renal cell carcinoma (ccRCC). RhoB favors neural crest formation.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
