General description

Ras-related protein Rab-21 (Rab21), a close homolog of Rab5, is ubiquitously expressed in human tissues. It localizes mainly to the early endocytic pathway. Rab21 is mapped to human chromosome 12q21.1 It belongs to the Rab family of small guanosine triphosphatases (GTPases). Rab proteins contain conserved regions, C-terminal hypervariable domains with a cysteine motif.

Specificity

Anti-Rab21 (C-terminal) recognizes human, rat, and mouse Rab21.

Immunogen

synthetic peptide corresponding to amino acid residues 203-216 of human RAB21, with an added cysteine, conjugated to KLH. The corresponding sequence differs by one amino acid in rat and mouse.

Application

Anti-Rab21 (C-terminal) antibody produced in rabbit may be used in:
  • immunoblotting
  • immunoprecipitation
  • immunofluorescence

Biochem/physiol Actions

Ras-related protein Rab-21 (Rab21) and Rab5 are integrin-associated proteins, acting as positive regulators of integrin traffic. The association of Rab21 with integrins modulates cell adhesion and migration. It is also involved in endosome morphology and function. Rab21 is associated with squamous carcinoma and its expression is elevated in glioma.

Physical form

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For extendedstorage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
