General description

Ribonucleic acid export 1 (RAE1/RNA export 1) also known as mRNA-binding protein.is a constituent of the mRNA export machinery. RAE1 has three protein/protein domains named β-transducin/WD40 motifs. The human RAE1 has a high homology to S.pombe Rae1 and Gle2 protein in S.cerevisiae. RAE1 gene is located on human chromosome 20q13.32.

Specificity

Anti-RAE1 (N-terminal) specifically recognizes human and rat RAE1 (43 kDa).

Immunogen

synthetic peptide corresponding to amino acids 18-35 of human RAE1, conjugated to KLH via a C-terminal cysteine residue. The corresponding sequence in mouse and rat differs by one amino acid.

Biochem/physiol Actions

Ribonucleic acid export 1 (RAE1/RNA export 1) participates in bipolar spindle formation through its interaction with nuclear mitotic apparatus protein (NuMA) and its binding to microtubules. RAE1 and nuclear pore complex protein (Nup98) form a complex with cdh1 and anaphase-promoting complex (APC) in early mitosis and specifically inhibit APC (Cdh1)-mediated ubiquitination of securin. It acts as a mitotic checkpoint regulator. It may act as a prognostic marker in breast cancers. Mutations in RAE1 leads in the accumulation of poly(A)-containing mRNA in the nucleus. It plays a key role in nucleoporin 98?kDa (NUP98)-mediated tumorigenesis in acute myeloid leukemia. Overexpression of this gene is observed in lung cancer.

Physical form

Solution in 0.01 M phosphate buffered saline pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8 °C for up to onemonth. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
