General description

Monoclonal Anti-PRMT3 (mouse IgG3 isotype) is derived from the PRMT3-367 hybridoma produced by the fusion of mouse myeloma cells (NS1) and splenocytes from BALB/c mice. Protein arginine methyltransferase 3 (PRMT3), one of the type I arginine methyltransferases is expressed in heart, small intestine, kidney, ovary, testis, thyroid, and cortex with a cytoplasmic localization within the cells. It consists of two important domains in its N-terminal region, a C2H2 zinc-finger motif and a tyrosine-phosphorylation consensus sequence. This gene is located on human chromosome 11p15.1.

Specificity

Does not cross-react with PRMT1, 2, 4, 5, or 6.

Immunogen

synthetic peptide corresponding to amino acids 501-514 of rat PRMT3 conjugated to KLH.

Monoclonal Anti-PRMT3 antibody produced in mouse may be used in:

  • immunocytochemistry
  • enzyme-linked immunosorbent assay (ELISA)

Biochem/physiol Actions

Protein arginine methyltransferase 3 (PRMT3) helps to catalyze the formation of asymmetric dimethylarginine. It plays a key role in ribosomal function and also in controlling protein synthesis. In apolipoprotein E (apoE) knockout mice, PRMT3 activity inhibition decreases hepatic steatosis. PRMT3 also serves as a specific cofactor for liver X receptor (LXR) transcription.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, and 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For prolonged storage, freeze in working aliquots at −20 °C. Repeated freezing and thawing is not recom-mended. Storage in frost-free freezers is also not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
