Parkin RBR E3 ubiquitin protein ligase (PARK2) is a putative tumor suppressor gene. It possesses an amino-terminal region containing the Ubl (ubiquitin like) domain and a carboxy-terminal region with four zinc coordinating domains, that is, RING0, RING1, in between ring (IBR) and RING2. Parkin is a 465 amino acid protein which is part of the ubiquitin-proteasome system that targets proteins for degradation. The gene encoding it is localized on human chromosome 6q25.2-q27.
Immunogen
synthetic peptide CI(298)KELHHFRILGEEQYN(313), corresponding to amino acid residues 298-313 from human parkin. The sequence is completely conserved between human, mouse, and rat.
Application
Anti-Parkin antibody produced in rabbit has been used in Western blotting and immunohistochemistry.
Biochem/physiol Actions
The expression of parkin RBR E3 ubiquitin protein ligase has been found to be altered in various malignancies. Mutations in the gene encoding it have been linked to autosomal recessive juvenile Parkinson (ARJP) disease. Parkin has a neuroprotective E3 ligase activity and polyubiquitylates proteins before degradation.
