Placenta growth factor (PlGF) is expressed in the placenta and somewhat less in the heart, lung and thyroid gland. Placentally expressed PlGF may act as an autocrine on trophoblasts, which express both PlGF and its receptor (VEGFR-1). Since these cells also make VEGF-A, natural heterodimers (PlGF/VEGF-A) have also been detected. Two alternatively spliced isoforms of PlGF have been identified.
Physical form
Lyophilized from a 0.2 μm filtered solution in 30% acetonitrile and 0.1% trifluoroacetic acid containing 50 μg bovine serum albumin per 1 μg of cytokine.
Preparation Note
Recombinant human PlGF is lyophilized from a 0.2 micron filtered solution of 30% CH3CN and 0.1% TFA (pH 7.4) containing 50 microg of bovine serum albumin per 1 microg of cytokine.
Reconstitution
Reconstitute with sterile PBS containing at least 0.1% HSA or BSA. The rhP/GF concentration should be no less than 10 μg/mL.
Analysis Note
The biological activity is measured by the ability to bind rhFlt-1/Fc in a functional ELISA. Immobilized rhFlt-1/Fc at 2 μg/mL (100 μL/well) can bind rhP/GF with a linear range at 0.1 - 5 ng/mL.
