General description

NEDD9 (neural precursor cell expressed, developmentally down-regulated 9) is a non-catalytic multi-domain scaffolding protein. The gene contains retinoic acid response element (RARE) element in its promoter region, and is composed of seven exons. It encodes for a protein composed of 834 amino acids in humans, and contains an N-terminal SH3 (src homology 3) domain, followed by SD - substrate domain, SRR - serine rich region, FAT - focal adhesion targeting domain. Its C-terminal contains a DYDY motif (amino acids 628–631). This gene is expressed ubiquitously, and shows strong expression in lungs and lung cell lines. NEDD9 is a member of the Cas (Crk-associated substrate) family of scaffolding proteins.
NEDD9 gene is mapped to human chromosome 6p24.2.

Anti-Nedd9 (C-terminal region) specifically recognizes human Nedd9.

Application

Anti-Nedd9 (C-terminal region) antibody produced in rabbit may be used in immunoblotting and immunofluorescence.

Biochem/physiol Actions

NEDD9 (neural precursor cell expressed, developmentally down-regulated 9) functions in the assembly of oncogenic kinase complexes such as focal adhesion kinase (FAK), ABL, SRC, Aurora-A (AURKA). It, therefore, controls signaling pathways implicated in processes such as cell cycle, migration, adhesion, invasion and chemotaxis that are involved in for tumorigenesis and metastases. In melanoma, glioblastoma, colon cancer and head and neck squamous carcinoma, the overexpression of this protein results in enhanced epithelial to mesenchymal transition, migration, and metastasis.

Physical form

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

Store at –20 °C. For continuous use, the product may be stored at 2–8 °C for up to one month. For extended storage, freeze in working aliquots at –20 °C. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
