General description

NUMB endocytic adaptor protein (Numb) is a conserved adapter protein. NUMB gene is mapped to human chromosome 14. The mammalian Numb gene gives rise to at least four alternatively spliced transcripts that produce four protein isoforms, Numb 1-4. Numb 1 and 3 have a proline-rich region.

Specificity

Anti-NUMB specifically recognizes human and rat NUMB.

Application

Anti-NUMB antibody may be used in immunoblotting.

Biochem/physiol Actions

Numb is an antagonist of the notch. It plays a role in the neural differentiation in the central nervous system (CNS) and self-renewal of neural stem cells (NSCs). Loss of Numb function in mice results in embryonic lethality and precocious neuronal differentiation. It has been suggested that a switch in Numb isoform expression is a critical step in cortical development. The proliferation of neural crest stem cells (NCSCs) is promoted by the proline-rich region (PRRL) of the isoform 1 and 3. The ectopic expression of Numb2 or Numb4 isoforms devoid of the proline?rich region mediates enhanced neuronal differentiation. The loss of Numb may impact the epithelial-to-mesenchymal transition (EMT).

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

Store at –20 °C. For continuous use, the product may be stored at 2–8 °C for up to one month. For extended storage, freeze in working aliquots at –20 °C. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.

Interstitial deletions of chromosome bands 14q24.1q24.3 are very rare with only three reported cases. We describe a 7-year-old boy with a 5.345 Mb de novo interstitial deletion at 14q24.1q24.3 band detected by array-CGH who had a complex phenotype characterized by seizures

Loss of numb function suggests that numb maintains progenitors in an undifferentiated state. Herein, we demonstrate that numb1 and numb3 are expressed in undifferentiated cortical progenitors, whereas numb2 and numb4 become prominent throughout differentiation. To further assess the role of
