Monoclonal Anti-Nck-2 (mouse IgG2b isotype) is derived from the 8.8 hybridoma produced by the fusion of mice myeloma cells and splenocytes from BALB/c mice immunized with a C-terminal region of human Nck-2 protein fused to glutathione S-transferase (GST). NCK adaptor protein 2 (Nck-2) is a 43 kDa protein, that is 68 % identical at the protein level as that of Nck-1.
Nck-2 is an adaptor protein that regulates signal transduction in cells. This protein consists of one C-terminal SH2 domain and three N-terminal SH3 domains. Nck-2 interacts with several growth factor signaling components such as EGFR, IRS-1 and PDGFR, and also with the integrin-linked kinase signaling protein, LINCH .
Specificity
The epitope recognized by the antibody resides within amino acids 176-274 of human Nck-2. Cross reactivity has been observed with human Nck-1. Monoclonal Anti-Nck-2 is specific for human, rabbit, rat and chicken Nck-2 (approx. 43 kDa). The product also binds to human Nck-1.
Immunogen
C-terminal region of human Nck-2 protein (amino acids 115-380) fused to GST
Application
Monoclonal Anti-Nck-2 antibody produced in mouse has been used in:
enzyme-linked immunosorbent assay (ELISA)
immunoblotting
immunoprecipitation
immunocytochemistry
Biochem/physiol Actions
NCK adaptor protein 2 (Nck-2) can interact with kinases like the focal adhesion kinase (FAK), which is a cytoplasmic tyrosine kinase involved in the cellular control of motility. It stimulates human melanoma cell proliferation.
Physical form
Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Many of the protein-protein interactions that are essential for eukaryotic intracellular signal transduction are mediated by protein binding modules including SH2, SH3, and LIM domains. Nck is a SH3- and SH2-containing adaptor protein implicated in coordinating various signaling pathways, including
