General description

Monoclonal Anti-Nedd8 (mouse IgG2b isotype) is derived from the ND23 hybridoma produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with a synthetic peptide corresponding to amino acids of human Nedd8 conjugated keyhole limpet hemocyanin (KLH). Nedd8 is a small protein (9 kDa) that shows the highest similarity to the ubiquitin protein (60% amino acid sequence identity).
Neural precursor cell expressed, developmentally down-regulated 8 (NEDD8) is an ubiquitin-like, 76 amino acid protein. It is expressed in endocrine and epithelial cells. NEDD8 belongs to the ubiquitin-like protein superfamily.

Immunogen

synthetic peptide corresponding to amino acids 20-32 of human Nedd8, conjugated to KLH.

Application

Monoclonal Anti-Nedd8 antibody produced in mouse has been used in:
  • enzyme-linked immunosorbent assay (ELISA)
  • immunoblotting
  • immunocytochemistry

Biochem/physiol Actions

Nedd8 conjugation has a physiological role in processes where the stability of nuclear factor kappa light chain enhancer of activated B cells (NF-κB) and p27 is important.
Neural precursor cell expressed, developmentally down-regulated 8 (NEDD8) is involved in pathways concerned with protein degradation. It targets substrates for neddylation, which is a post-translational modification. NEDD8 also regulates the signaling of proteins like cullins.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Recent studies indicate that post-translational protein neddylation is required for the maintenance of cell viability in several lymphoma cell lines, while inhibition of the neddylation pathway with an NEDD8-activating enzyme (NAE) inhibitor MLN4924 induces apoptosis in lymphoma cells.
