General description

Mammalian calpains are calcium-activated, neutral, non-lysosomal, intracellular proteases that may be ubiquitous, stomach-specific, or muscle-specific. Calpain 1 is a ubiquitous protease that is involved in calcium homeostasis and the regulation of apoptosis. In cardiomyocytes, activated calpain 1 induces apoptosis by BID cleavage and release of cytochrome c from the mitochondria.

The Human Calpain 1 Inhibitor Screening Kit is a rapid, simple and sensitive assay that is suitable for high-throughput screening of Calpain 1 inhibitors. Calpain 1 activity is measured by cleaving a synthetic 7-amino-4-trifluoromethylcoumarin (AFC)-based substrate (Ac-LLY-AFC) to yield AFC, a fluorescent product (λex = 400/λem = 505 nm), proportional to the enzymatic activity present.

Principle
