The use of RIPA or other harsh cell lysis buffers is not recommended, as they may denature and/or inhibit enzymes that are crucial for the kit's performance. According to the protocol of MAK207, tissue samples (10 mg) can be homogenized in 100 mL of ice-cold G3P Assay Buffer (MAK207A in the kit), kept on ice for 10 minutes, centrifuged at 12,000 x g for 5 minutes to remove insoluble material, and then transferred to a fresh tube.
