General description

α-Ketoglutarate (α-KG) is a key intermediate in the Tricarboxylic Acid (TCA) Cycle, formed by the oxidative decarboylation of isocitrate by isocitrate dehydrogenase. The synthesis of α-KG from glutamate by glutamate dehydrogenase is a key TCA cycle anaplerotic reaction. α-KG is an important nitrogen transporter. This α-Ketoglutarate Assay Kit provides a simple, sensitive and rapid means for quantifying α-KG in a variety of samples.

Suitability

Suitable for the measurement of a-Ketoglutarate in a variety of biological samples including cell and tissue lysates

Principle

α-KG concentration is determined by a coupled enzyme assay, which results in a colorimetric (570 nm)/ fluorometric (λex = 535/λem = 587 nm) product, proportional to the α-KG present. Typical sensitivities of detection for this kit are 0.01 to 10 nmoles.

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature.   2. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
      For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  3. Is a 10KDa MWCO spin filter a suitable substitute for perchloric acid in the deproteinization step of the Acetyl-CoA Assay Kit (MAK039)?

    1. Perchloric acid (PCA) is recommended for deproteinization, but if unavailable, 10 KDa spin filters can be used as an alternative.

    1. This kit would be suitable for plant samples assuming that the concentration of a-Ketoglutarate present in the tissue lysate is sufficient to enable detection. This α-Ketoglutarate Assay Kit is suitable for the measurement of a-Ketoglutarate in biological samples including cell and tissue lysates.
