General description

Livin or baculoviral IAP repeat-containing 7 (BIRC7) belongs to the family of the inhibitor of apoptosis proteins (IAP). It contains a single copy of a baculovirus IAP repeat (BIR) as well as a really interesting new gene (RING)-type zinc finger domain. Structurally, livin comprises a three-stranded anti-parallel β-sheet and four α-helices and takes a globular architecture. BIRC7 gene is mapped to human chromosome 20q13.33 and is expressed in α and β isoforms.

Anti-Livin (C-terminal) recognizes human Livin/BIRC7.

Biochem/physiol Actions

Livin or baculoviral IAP repeat-containing 7 (BIRC7) inhibitory functionality is mediated by the baculovirus inhibitor of apoptosis proteins (IAP)repeat (BIR) domain. This domain is also essential for interaction with caspases. The really interesting new gene (RING) finger domain elicits E3-ubiquitin ligase activity in the absence of BIR domain. The two isoforms of livin have different antiapoptotic properties, with isoform protecting cells from apoptosis induced by staurosporine and isoform α protecting cells from apoptosis induced by etoposide. An elevated expression of Livin is detected in prostate, breast, colon, head and neck squamous cell carcinoma. Inhibition of livin in nasopharyngeal carcinoma (NPC) leads to improved irradiation sensitivity in NPC cells.

Physical form

Solution in 0.01 M phosphate buffered saline pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2–8 °C for up to one month. For extended storage freeze in working aliquots. Repeated freezing and thawing is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
