General description

Low-density lipoprotein receptor-related protein 6 (LRP6) belongs to the low-density lipoprotein receptor (LDLR) family and is expressed majorly in the nervous system. It is closely related to LRP5. LRP6 gene is mapped to human chromosome 12p13.2.

Anti-LRP6 specifically recognizes human LRP6.

Application

Anti-LRP6 antibody produced in rabbit may be used in immunoblotting and immunoprecipitation.

Biochem/physiol Actions

Low-density lipoprotein receptor related protein 6 (LRP6) is part of the wingless (Wnt)-frizzled-LRP6 complex, that recruits axin to the plasma membrane. This results in the inhibition of beta-catenin phosphorylation and subsequent degradation and activation of T-cell factor/lymphoid enhancer-binding factor 1 (TCF/LEF-1)-dependent transcription. LRP6 is essential for proper brain development. Deletion of the LRP6 gene results in severe brain abnormalities. Mutations in LRP6 are associated with coronary heart disease and osteoporosis. LRP6 has been shown to undergo phosphorylation at multiple PPPSP motifs upon stimulation by Wnt. The phosphorylation of LRP6 by glycogen synthase kinase 3 (GSK3) and casein kinase 1 (CKI) is induced by Wnt. Regulated proteolytic processing of LRP6 has shown to release its intracellular domain (ICD). This fragment binds to GSK3 and inhibits its activity, resulting in activation of the canonical Wnt signaling pathway. A higher expression of LRP6 is detected in tumors.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

Store at –20 °C. For continuous use, the product may be stored at 2–8 °C for up to one month. For extended storage, freeze in working aliquots at –20 °C. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
