indirect immunofluorescence: 0.5-1 μg/mL using paraformaldehyde fixed HEK-293T cells over expressing human PLAGL1 western blot: 1-2 μg/mL using lysates of HEK-293T cells over expressing human PLAGL1
The gene PLAGL1 (pleiomorphic adenoma-like protein 1), PLAGL2 and PLAG1 form a subfamily of PLAG family of zinc finger proteins that recognize DNA and RNA. The three members share similarity at the amino-terminal, but are functionally different, varying in their DNA binding capacities. The gene is mapped to human chromosome 6q24.2, a region implicated in several cancers.
Specificity
Anti-PLAGL1 (C-terminal) specifically recognizes human PLAGL1.
Application
Anti-PLAGL1 (C-terminal) antibody produced in rabbit may be used in immunoblotting and immunofluorescence.
Biochem/physiol Actions
PLAGL1 gene expression is downregulated in a methylation-sensitive manner. PLAGL1 is an imprinted gene that mediates an imprinted gene network that plays an important role in embryonic growth control.
The maternally-imprinted gene, PLAGL1 (pleiomorphic adenoma-like protein 1), functions as an oncogene as well as a tumor suppressor depending on the cell context. It has been found to exhibit anti-proliferative properties and regulate apoptosis and cell-cycle arrest via p53.
Physical form
Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
Storage and Stability
Store at –20 °C. For continuous use, the product may be stored at 2–8 °C for up to one month. For extended storage, freeze in working aliquots at –20 °C. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
Disclaimer
Unless otherwise stated in our catalog or other company documentation accompanying the product(s), our products are intended for research use only and are not to be used for any other purpose, which includes but is not limited to, unauthorized commercial uses, in vitro diagnostic uses, ex vivo or in vivo therapeutic uses or any type of consumption or application to humans or animals.
Soft tissue sarcomas (STS) are rare, complex tumors with a poor prognosis. The identification of new prognostic biomarkers is needed to improve patient management. Our aim was to determine the methylation status of the 118 CpG sites in the PLAGL1
