General description

JAB1 is a highly conserved protein expressed in mammalian tissues. This protein is found both in the nucleus and the cytoplasm.
The gene JAB1 (Jun activation domain-binding protein-1) encodes a component of COP9 signalosome that functions in the phosphorylation of specific transcription factors. The gene is mapped to human chromosome 8q13.2. The encoded protein contains 334 amino acids with a molar mass of 38kDa.

Specificity

Whole antiserum is purified to provide an IgG fraction of antiserum Anti-JAB1 (SV-16) recognizes human, mouse, and rat JAB1.

Immunogen

synthetic peptide corresponding to amino acid residues 201-216 of human JAB1, conjugated to KLH. The corresponding sequence is identical in mouse and rat.

Anti-JAB1 (SV-16) antibody produced in rabbit may be used in:

Biochem/physiol Actions

The gene JAB1 (Jun activation domain-binding protein-1), also referred to as COPS5 (COP9 subunit 5), is a coactivator of AP-1 (activator protein 1) transcription factor. It is also involved in the regulation of cell cycle by degradation of the cyclin-dependent kinase inhibitor p27Kip1. It increases the levels of endogenous phospho-c-Jun by activating c-Jun amino-terminal kinase (JNK) activity. These activities are inhibited by association with macrophage migration inhibitory factor (MIF), a critical cytokine in several inflammatory responses. This binding, however, does not affect induction of the transcription factor NF (nuclear factor)-κB. It exists as a monomer and functions in deneddylation of NEDD8 (neural precursor cell expressed, developmentally down-regulated 8). It facilitates the shuttling of certain molecules between the nucleus and cytoplasm. JAB1 regulates cellular proliferation and apoptosis by modulating intracellular signaling. Overexpression of this protein has been associated with tumorigenic process.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

Store at -20 °C20 °C. For continuous use, store at 2-8 °C for up to one month. For prolonged storage, freeze in working aliquots at-20 °C. Repeated freezing and thawing, or storage in frost-free freezers,is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
