Gene Information
mouse ... Dffa(13347)
General description
The gene DFF-45, also referred to as DFFA (DNA fragmentation factor subunit α), encodes the α subunit of the heterodimeric protein DFF (DNA fragmentation factor) that also consists of the β subunit encoded by DFFB. The molar mass of the protein encoded by DFFA is 45kDa. The gene is mapped to human chromosome 1p36.3-p36.2.
Immunogen
synthetic peptide corresponding to amino acids 312-331 of mouse DFF45/ICAD (DNA Fragmentation Factor 45/ Inhibitor of Caspase-3 Activated DNase I).
Biochem/physiol Actions
The protein encoded by the gene DFFA (DNA fragmentation factor subunit α), also referred to as ICAD (inhibitor of caspase-activated deoxyribonuclease), serves as a substrate for caspase-3. It is cleaved at two sites by caspase-3 in response to apoptotic signals, and DFF is activated when DFFA separates from DFFB, the active nuclease that is also referred to as CAD (caspase-activated deoxyribonuclease). DFFA functions as a chaperone facilitating the correct folding of DFFB, and also as an inhibitor of DFFB. Active DFFB functions in the fragmentation of DNA by introducing double-stranded breaks during apoptosis.

CAD/DFF40, the nuclease responsible for DNA fragmentation during apoptosis, exists as a heterodimeric complex with DFF45/ICAD. This study determines the molecular mechanisms of regulation of DFF40 via the chaperone and inhibition activities of DFF45. We analyze proteins corresponding to the

The homeostasis of animals is regulated not only by the growth and differentiation of cells, but also by cell death through a process known as apoptosis. Apoptosis is mediated by members of the caspase family of proteases, and eventually causes

We have identified and purified from HeLa cytosol a protein that induces DNA fragmentation in coincubated nuclei after it is activated by caspase-3. This protein, designated DNA Fragmentation Factor (DFF), is a heterodimer of 40 kDa and 45 kDa subunits.
