General description

The gene DFF-45, also referred to as DFFA (DNA fragmentation factor subunit α), encodes the α subunit of the heterodimeric protein DFF (DNA fragmentation factor) that also consists of the β subunit encoded by DFFB. The molar mass of the protein encoded by DFFA is 45kDa. The gene is mapped to human chromosome 1p36.3-p36.2.

Biochem/physiol Actions

The protein encoded by the gene DFFA (DNA fragmentation factor subunit α), also referred to as ICAD (inhibitor of caspase-activated deoxyribonuclease), serves as a substrate for caspase-3. It is cleaved at two sites by caspase-3 in response to apoptotic signals, and DFF is activated when DFFA separates from DFFB, the active nuclease that is also referred to as CAD (caspase-activated deoxyribonuclease). DFFA functions as a chaperone facilitating the correct folding of DFFB, and also as an inhibitor of DFFB. Active DFFB functions in the fragmentation of DNA by introducing double-stranded breaks during apoptosis.
