General description

The gene RPS14 (40S ribosomal protein S14) has been mapped to human chromosome 5q.

Application

Biochem/physiol Actions

40S ribosomal protein S14 (RPS14) binds and inactivates E3-ubiquitin ligase (MDM2), thereby leading to p53-dependent cell-cycle arrest and growth inhibition. In addition, RPS14 inhibits c-Myc transcriptional activity, causing suppression of c-Myc-induced cell proliferation. The 5q-syndrome, a subtype of myelodysplastic syndrome. The RPS14 gene shows haploinsufficiency in the CD34+ cells of patients with this syndrome. Haploinsufficiency of RPS14 results in a block in erythroid differentiation. RPS14 is down-regulated in bone marrow derived CD34+ cells from patients with non-5q-myelodysplastic syndrome. Lower RPS14 expression is associated with increased apoptosis of nucleated erythrocytes.
