Gene Information

human ... H4-16(121504)

General description

The building block of chromatin is the nucleosome which is comprised of an octamer of core histones (H2A, H2B, H3, and H4) around which 147 bp of DNA are wrapped. Histones H3 and H4 are the predominant histones subjected to covalent modifications.[1][2] Acetylation of histone H4 on Lys16 (H4-K16Ac) is a prevalent modification that plays a role in transcriptional activation by regulating higher-order chromatin structure and protein interactions.[3][4]

Immunogen

synthetic peptide corresponding to amino acids 9-18 (Ac-Lys16) of human histone H4, conjugated to KLH.

Application

Anti-acetyl-Histone H4 (Ac-Lys16) antibody produced in rabbit is suitable for western blot at a concentration of 1-2μg/mL using HL60 cell lysates.
Applications in which this antibody has been used successfully, and the associated peer-reviewed papers, are given below.
Chromatin immunoprecipitation (1 paper)
Immunohistochemistry (1 paper)

Target description

acetyl-Histone H4 (Ac-Lys16) is intronless and encodes a member of the histone H4 family. Transcripts from this gene lack polyA tails; instead, they contain a palindromic termination element.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

The Sir3 protein helps form telomeric heterochromatin by interacting with hypoacetylated histone H4 lysine 16 (H4-Lys16). The molecular nature of the heterochromatin boundary is still unknown. Here we show that the MYST-like acetyltransferase Sas2p is required for the acetylation (Ac)

Histone post-translational modification heritably regulates gene expression involved in most cellular biological processes. Experimental studies suggest that alteration of histone modifications affects gene expression by changing chromatin structure, causing various cellular responses to environmental influences.
