Gene Information

human ... HUS1(3364)

General description

HUS1 checkpoint protein is a member of the genotoxin-activated checkpoint (Hus1-Rad1-Rad9) complex. The HUS1 gene is mapped on the human chromosome at 7p12.3.

Specificity

Anti-HUS1 specifically recognizes human HUS1.

Immunogen

synthetic peptide corresponding to amino acids 147-164 of human HUS1, conjugated to KLH via an N-teminal added cysteine residue.

Application

Anti-HUS1 antibody produced in rabbit may be used in immunoblotting and immunoprecipitation.

Biochem/physiol Actions

HUS1 checkpoint protein is involved in DNA repair and cell cycle arrest as a result of DNA damage. Upregulation of HUS1 gene expression is associated with the inhibition of cell proliferation, migration, invasion, and cell cycle arrest in hepatocellular carcinoma (HCC). Loss of HUS1 gene leads to embryonic lethality.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For extended storage, freeze in working aliquots. Repeated freezing and thawing, or storage in “frost-free” freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.

The Rad9-Rad1-Hus1 (9-1-1) cell cycle checkpoint complex plays a key role in the DNA damage response. Cells with a defective 9-1-1 complex have been shown to be sensitive to apoptosis induced by certain types of genotoxic stress. However, the mechanism
