General description

HIPK1 (homeodomain-interacting protein kinase 1) is a serine/threonine protein kinase. It belongs to the HIPK family. The protein is present in the nucleus.[1]

Biochem/physiol Actions

HIPK1 (homeodomain-interacting protein kinase 1) is responsible for phosphorylation of homeodomain transcription factors. HIPK1 interacts with DAXX (death domain-associated protein 6) and decreases its transcriptional repression activity.[1] It is up-regulated in breast cancer cell lines and is suggested to be involved in tumorigenesis, partly by serine phosphorylation of p53.[2]

Physical form

Supplied in 50 mM Tris-HCl, pH 7.5, with 150 mM NaCl, 0.2 5mM DTT, 0.1 mM EGTA, 0.1 mM EDTA, 0.1 mM PMSF, and 25% glycerol.

The tumor suppressor p53 is regulated in part by binding to cellular proteins. We used p53 as bait in the yeast two-hybrid system and isolated homeodomain-interacting protein kinase 1 (HIPK1) as a p53-binding protein. Deletion analysis showed that amino acids

We describe an interaction between homeodomain-interacting protein kinase 1 (HIPK1) and Daxx, two transcriptional regulators important in transducing growth-regulatory signals. We demonstrate that HIPK1 is ubiquitously expressed in mice and humans and localizes predominantly to the nucleus. Daxx normally resides
