Biochem/physiol Actions

Peptidyl-prolyl cis-trans isomerase NIMA-interacting 1 is an enzyme encoded by the PIN1 gene in humans and belongs to parvulin PPIase family. It is an essential PPIase (peptidyl-prolyl cis/trans isomerase) that regulates mitosis presumably by interacting with NIMA and attenuating its mitosis-promoting activity. Pin1 regulates proteins involved in cell proliferation and pluripotency or cellular invasion. It is overexpressed in several human cancers and contributes to tumorigenesis. Pin1 plays a crutial role in cell transformation and oncogenesis. It is necessary for the regulation of the enzyme as well.

PIN1 is one member of the parvulin PPIase family. By controlling Pro-directed phosphorylation, PIN1 plays an important role in cell transformation and oncogenesis. There are many polymorphisms in the PIN1 gene, including rs2233678 and rs2233679 affecting the PIN1 promoter. Recently

Post-translational modifications are critical to modulate protein function. A post-translational mechanism, peptidyl prolyl cis-trans isomerisation, plays a key role in protein regulation. Pin1 is a ubiquitous peptidyl prolyl cis-trans isomerase conserved from Archae to Human. This enzyme binds and isomerizes

Enzymes catalyze a plethora of chemical reactions that are tightly regulated and intricately coupled in biology. Catalysis of phosphorylation-dependent cis-trans isomerization of peptidyl-prolyl bonds, which act as conformational switches in regulating many post-phosphorylation processes, is considered to be one of

The NIMA kinase is essential for progression through mitosis in Aspergillus nidulans, and there is evidence for a similar pathway in other eukaryotic cells. Here we describe the human protein Pin1, a peptidyl-prolyl cis/trans isomerase (PPIase) that interacts with NIMA.
