Biochem/physiol Actions

Vesicle-associated membrane protein 2 is a small integral membrane protein encoded by the VAMP2 gene in humans and is mapped on chromosome 17. It is a key component of the synaptic vesicle docking/fusion machinery and its mRNA. VAMP2 may be involved in the mechanism of action of antidepressants and may alter their efficacy. It is unlikely to play a major role in the efficacy of fluvoxamine. VAMP2 is recognized and cleaved by BoNT/B (Botulinum neurotoxin) and TeNT (tetanus neurotoxin) that possess similar organization.

Botulinum neurotoxins (BoNTs), the most potent toxins known, are potential bioterrorism agents. It is well established that all seven serotypes of BoNTs (BoNT/A-G) require complex gangliosides as co-receptors. Here, we report that BoNT/DC, a presumed mosaic toxin between BoNT/D and

Botulinum neurotoxin (BoNT; serotypes A-G) and tetanus neurotoxin elicit flaccid and spastic paralysis, respectively. These neurotoxins are zinc proteases that cleave SNARE proteins to inhibit synaptic vesicle fusion to the plasma membrane. Although BoNT/B and tetanus neurotoxin (TeNT) cleave VAMP-2

Dynamin is a multidomain GTPase exhibiting mechanochemical and catalytic properties involved in vesicle scission from the plasmalemma during endocytosis. New evidence indicates that dynamin is also involved in exocytotic release of catecholamines, suggesting the existence of a dynamin-regulated structure that
