General description
Application
The assay is based on the competition between unlabeled thromboxane B2 and a fixed quantity of peroxidase-labeled thromboxane B2 for a limited number of binding sites on a thromboxane B2-specific antibody. With fixed amounts of antibody and peroxidase-labeled thromboxane B2, the amount of peroxidase- labeled ligand bound by the antibody will be inversely proportional to the concentration of added unlabeled ligand. The peroxidase ligand that is bound to the antibody is immobilized on to polystyrene microplate wells precoated with second antibody. Therefore, any unbound ligand can be removed from the well by simple washing procedures. The amount of peroxidase labelled thromboxane B2 bound to the antibody is determined by addition of a tetramethylbenzidine (TMB)/ hydrogen peroxide single pot substrate. The reaction is stopped by additon of an acid solution, and the resultant colour read at 450 nm in a microplate photometer. The concentration of unlabelled thromboxane B2 in a sample is determined by interpolation from a standard curve.
