Immunogen

synthetic peptide corresponding to the third extracellular loop of human frizzled-2, conjugated to KLH. The immunizing peptide has 100% homology with rat and mouse gene.

Application

Anti-Frizzled-2 antibody is suitable for western blotting and immunofluorescence labelling to study the expression of Wnt5a and its receptor Fzd2 in the spinal cord of adult amyotrophic lateral sclerosis transgenic mice. It is also suitable for immunohistochemistry studies formalin-fixed, paraffin-embedded sections in concentration of ~7.6μg/mL, using human kidney, fetal.

Biochem/physiol Actions

Frizzled-2 belongs to frizzled (FZD) family. Frizzled homolog protein (FZD) is a seven-pass transmembrane type receptor expressed in various normal tissues (skeletal muscle, kidney, pancreas, cerebellum and cerebral cortex). In human, ten members (FZD1-FZD10) of Frizzled homolog protein have been identified. Generally, it is expressed in most human adult and fetal tissues, but it is found to be over expressed in several cancers such as primary Wilms′ tumour, melanoma and lung squamous cell carcinoma. FZD2 is directly associated with Wnt signaling pathway. It binds to Wnt5a to activate the WNT/Ca2+signaling pathway in melanoma cell lines and also in presence of Wnt3a, can facilitate Wnt/β-catenin signaling in pulmonary carcinoma.
