Biochem/physiol Actions

Filamin, also known as actin-binding protein, is a large flexible V-shaped actin cross-linking protein present in vertebrates of both muscle and non-muscle cells. It has actin-binding domains at the ends of each arm. Filamin is a dimer of two large (280-kd) subunits, forming a flexible V-shaped molecule that crosslinks actin filaments into orthogonal networks. Filamin cross-links actin and promotes it′s aggregation and gelation.

Chicken gizzard filamin has been digested with purified Ca2+-activated protease. The subunits of (Mr = 250,000) of the protein are cleaved asymmetrically into two fragments, heavy merofilamin, Mr = 240,000, and light merofilamin, Mr = 9,500. Digestion is complete at

Calpain represents a family of Ca(2+)-dependent cytosolic cysteine proteases found in almost all eukaryotes and some bacteria, and is involved in a variety of biological phenomena, including brain function. Several substrates of calpain are aggressively proteolyzed under pathological conditions, e.g.
