Cell Line Description

The hybridoma line S2 has been obtained by fusion of the myeloma P3x63 Ag6.653 and spleen cells from (C57 BL/6 x BALB/c) F1 female mice after a single i.p. injection of lymphoblastoid cells. These lymphoblastoid cells were DRw6 homozygous and expressed HLA A2, Bw44, CW5, DRw6 and 16.23+. The monoclonal antibody secreted by S2 defines an epitope apparently restricted to DRQ13. Together with antibodies expressed by 16/23 (Sigma Catalogue number. 99043001) and S5 (Sigma Catalogue number. 99061827) they are useful tools for the biochemical dissection of DRw6 related epitopes. S2 has been analysed in International Histocompatibility Workshop (IOWS 3021, II WS 0826). The cells have been eradicated from mycoplasma before depositing at ECACC. The cells have a doubling time of about 36 hours.

RPMI 1640 + 2mM glutamine + 10% FBS

Subculture Routine

Maintain cultures between 3-9 x 100,000 cells/ml, 5% CO2; 37°C.
