Cell Line Origin

Hamster Chinese ovary (transfected with rat m1 mAChR encoding gene).

Cell Line Description

Derived in 1990 by transfecting Chinese Hamster ovary CHO-K1 cells with the gene encoding rat m1 muscarinic acetylcholine receptor (mAChR) in the mammalian expression vector, pSVL. These cells were then co-transfected with pMSVNeo and selected for growth in selection medium containing 500μg/ml G418. The cells are reported to express 800 fmol of receptor/mg of membrane protein.

Application

This cell line can be used to study m1 mAChR in cells devoid of other muscarinic receptor subtypes.

Culture Medium

Ham′s F12 + 2mM Glutamine + 50-100μg/ml G418 + 10% Foetal Bovine Serum (FBS).

Subculture Routine

Split sub-confluent cultures (70-80%) 1:4 to 1:8 i.e. seeding at 1-2x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C.
