Human pancreatic adenocarcinoma

Cell Line Description

The human pancreatic cancer cell line PSN1 was derived from pancreatic adenocarcinoma tissue. After transplantation into nude mice the PSN1 cell line was established from the xenograft. PSN1 cells are unique in that amplification of both c-myc (50-fold) and activated c-Ki-ras (3-6 fold) with a point mutation from GGT to CGT at codon 12 are present in the same degree as in the original tumour and contain increased amounts of c-myc and c-Ki-ras transcripts. Loss of one of the two p53 alleles and a point mutation from AAG to CAG at codon 132 in the remaining allele was also observed in PSN1.

Application

PSN1 has been used to study synthetic acetylhomoagmatine in vitro. It has also been used to determine the cytotoxicity of irinotecan and topotecan.
PSN1 is an example of a cancer cell having multiple genetic changes including those of two oncogenes (c-Ki-ras and c-myc) and a tumour suppresser gene (p53).

Culture Medium

RPMI 1640 + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).

Subculture Routine

Split sub-confluent cultures (70-80%) 1:3 to 1:8 i.e. seeding at 1-3 x 10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C.
