Cell Line Origin

Rat embryonic fibroblasts RSV-33 transformed

Cell Line Description

The R74 line was derived by co-cultivation of 18 day-old Lewis rat embryonic fibroblasts with cells of a chicken positive for Rous sarcoma virus-33 (RSV-33). The R9 cell population is clearly distinguishable from RT1 and R74 cell lines because it contains flat polygonal and triangular refractile cells with a tendency to round up. It is also tumourigenic in chickens and rats.

Culture Medium

EMEM (EBSS) + 2mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 5% Foetal Bovine Serum (FBS).

Subculture Routine

Split sub-confluent cultures (70-80%) 1:2 to 1:4 i.e. seeding at 2-4x10,000 cells/cm2 using 0.25% trypsin/EDTA; 5% CO2; 37°C.
