Cell Line Description

This cell line has the capacity to be activated to produce murine leukaemia virus (MLV). Mice of the high leukaemic inbred strain AKR regularly contain high levels of MLV throughout most of their post-natal life. However, studies of the AKR mouse embryo indicated that they contain very little, or no, infectious virus. Cell lines were found to produce virus only after several weeks in culture. Virus-negative cell lines can be induced to initiate synthesis of MLV . Multiple transforming growth factors (TGFs) have been isolated from the serum-free conditioned media and acid-ethanol cell extracts of non-transformed AKR-2B cells.

Culture Medium

McCoy′s 5a + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).

Subculture Routine

Split sub-confluent cultures (70-80%) 1:16 to 1:24 i.e. seeding at 4-10x1,000 cells/cm2 using 0.25% trypsin/EDTA; 5% CO2; 37°C. Do not allow to reach confluence or grow over 20 passages.
