From a cervical carcinoma of a 41 year old Caucasian female. Cells form spreading predominantly monolayered, epithelial colonies with hemicysts which are characteristic of transporting epithelia and respond to crowding by separation and shedding. These cells have retained properties of basal cell differentiation in long term culture and exhibit G6PD type B. C-4 II cells have been reported to contain human papilloma virus 18 (HPV-18) DNA sequences and to express HPV-18 RNA.
Split sub-confluent cultures (70-80%) 1:4 to 1:10 i.e. seeding at 1-3x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C.
