A temperature sensitive mutant isolated from the mouse mammary cell carcinoma cell line, FM3A (Sigma Catalogue number. 87100804). Cells will not divide at 39°C (non-permissive temperature) and will grow at 33°C (permissive temperature). Electron microscope findings have shown that chromatin abnormally condenses into fragmented and compact forms in 80% of the cells at 39°C for 16 hours. This abnormal condensation does not occur at 33°C or in the FM3A cell line at either temperature.
Maintain cultures between 1-3x100,000 cells/ml; 5% CO2; 33°C.
