Adherent keratinocyte cell line derived from a lymph node metastasis squamous cell carcinoma (SSC) of the larynx. Keratin and involucrin markers present. Known mutations: p53, p16 and p14ARF. Cells are tumourigenic in athymic mice. By short tandem repeat (STR)-PCR analysis the Y chromosome could not be detected in this cell line. It is a known phenomenon that SSC cell lines can lose their Y chromosome.

Subculture Routine

Split sub-confluent cultures (70-80%) 1:2 using 0.05% trypsin/EDTA; 8% CO2; 37 C. Suggested seeding density 4 x 10,000 cells/cm2. A feeder layer of lethally-irradiated or mitomycin C-treated 3T3 feeder cells can be used to culture this cell line, howeve
