Application

Kinase activity is measured as the molar amount of phosphate incorporated into the CHKtide per minute per mg protein at 30 °C using a final concentration of 50 μM [32P] ATP.

Biochem/physiol Actions

Chk2 is the mammalian homolog of the S.cerevisiae Rad53 and S.pombe Cds1 protein kinase required for DNA damage and replication checkpoints. Chk2 is rapidly activated, by phosphorylation, in response to replication blocks and DNA damage.

Unit Definition

One unit will incorporate 1 nanomole of phosphate into CHKTIDE substrate per minute at pH 7.2 at 30 deg C.
