General description

c-Raf or Raf-1 belongs to the Raf family of kinases that have regulatory functions in cell growth, differentiation, growth factor receptor stimulation via the MAPK signalling. The other members of the family are A-Raf and B-Raf. All the isoforms have unique as well as overlapping regulatory functions. c-Raf kinase is the first identified downstream effector kinase of Ras, and is the activator of MEK which in turn activates ERK. Phosphorylation of c-Raf at Ser338 and Tyr340 regulates its biological function.
Anti-phospho-c-Raf [pSer338/pTyr340] detects human c-Raf [pSer338/pTyr340]hosphorylated at [pSer338/pTyr340]er338 and [pSer338/pTyr340]yr340 (74 kDa). Mouse and [pSer338/pTyr340]at c-Raf (100% homology) are [pSer338/pTyr340]xpected [pSer338/pTyr340]o cross react.

Immunogen

synthetic phosphopeptide derived from the region of human c-Raf that contains serine 338 and tyrosine 340.

Application

The antibody may be used to detect phospho-c-Raf (Ser338/Tyr340) by immunoblotting at working dilution of 1:1000 in cell lysates from HEK293 cells transfected with c-Raf. Detection was also possible in spermatozoa of mice at 1:1000 dilution.

Physical form

Solution in Dulbecco′s phosphate buffered saline (without Mg2+ and Ca2+), pH 7.3, with 50% glycerol, 1.0 mg/mL BSA (IgG and protease free) and 0.05% sodium azide.
