Specificity

Recognizes the CD45RO human cell surface glycoprotein. The epitope recognized by this antibody is sensitive to neuraminidase digestion, but not to formalin fixation and paraffin embedding. The antibody is capable of detecting CD45RO antigen in tissue sections processed with a variety of fixatives. The antibody also detects CD45RO antigen in frozen sections, cell suspension, smears, imprints and cytospins.

Application

Monoclonal Anti-CD45RO antibody produced in mouse is suitable for flow cytometry at a dilution of 10μL using 1 × 106 cells.

Biochem/physiol Actions

Protein tyrosine phosphatase, receptor type, C, known as PTPRC, is an enzyme encoded by the PTPRC gene in humans. CD45RO is N-glycosylated. Deglycosylation at N327 of CD45RO results in increased binding to galectin-3 without affecting apoptosis, while deglycosylation at N36 or N109 of CD45RO enhanced galectin-3-induced apoptosis.[1] The low levels of CD45 expression are specific to chronic lymphocytic leukemia (CLL) cells and reflect cell fragility.[2]

Target description

CD45RO is a single-chain transmembranous glycoprotein, which represents the low molecular weight isoform of the leukocyte common antigen (LCA). It is expressed on most thymocytes, about 45% of resting peripheral blood T cells, virtually all T cells in skin reactive infiltrates and the majority of T cell malignancies. It is also found on a subset of B cells and on some B cell lymphomas. NK-cells do not express this antigen.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 1% bovine serum albumin and 15 mM sodium azide.
